Population Genetics Robarts, Daniel [1], Wolfe, Andrea [1]. Analysis of genetic structure of Viola pedata (Violaceae) via microsatellite markers indicates multiple Pleistocene refugia. Viola pedata L. (Violaceae), a unique, and broadly distributed, polyploid species, using eight microsatellite loci. In an analysis of more than 450 individuals collected from 43 populations widely distributed across the breadth of its range, we provide the first large-scale genetic profile of a North American representative of Viola. Loci reviewed were inconsistently variable, demonstrating as many as 20 or as few as five alleles, and analysis uncovered regionally unique alleles. Analysis by STRUCTURE indicated six major clusters, while AMOVA by population and cluster indicate these criteria combined to contribute over 20% of the genetic variation. Detected genetic clusters correspond to distinct regional lineages, and location of the grouped populations suggests this species persisted in multiple northern refugia during previous glacial maxima.
