Conservation of the Auxin Response Factor (ARF) MONOPTEROS (MP) gene lineage in flowering plants.

The MONOPTEROS (MP) gene encodes AUXIN RESPONSE FACTOR5 (ARF5) that activates auxin-responsive target genes in the model eudicot, Arabidopsis thaliana. Although well characterized in A. thaliana, little is known about MP orthologs in other plant taxa. Bayesian phylogenetic analyses estimate a well-supported MP clade comprised of monocots and eudicots indicating a origin for the gene family near the base of the flowering plant clade, and potentially deeper within vascular plants. Comparative genomic analysis of Arabidopsis thaliana, Brachypodium distachyon, Oryza sativa, Mimulus guttatus, and Cucumis sativus reside in syntenic chromosomal regions supporting the results of the phylogenetic analysis. Comparative mRNA expression analyses using quantitative RT-PCR and in situ mRNA hybridization reveal conserved expression patterns in axillary meristems and vascular tissue in the grasses Brachypodium distachyon and Oryza sativa and the eudicots Mimulus guttatus (Phyrmaceae), Carica papaya (Caricaceae), and Cucumis sativus (Cucurbitaceae). Together, these analyses point to a conserved role for MP co-orthologs in both monocots and eudicots.
